Roughly 280 Spider-People Show Up on Screen
The Spider-Society scenes are absolutely rammed with alternate Spider-heroes, somewhere around 280 of them by most counts, though only about a third actually get a name and a line of dialogue. The rest are just background gags for eagle-eyed fans to freeze-frame and argue about online.
The Joker Writer Snuck in a Lost Tribute
Mark Hamill is apparently a massive Lost fan, and lead writer Paul Dini actually wrote for the show, so a few nods snuck their way into the script. Near the end of the game there's a short speech from 'Joker' (really Clayface wearing his face) that echoes the tone of Lost's series finale pretty closely if you know what you're listening for.

The School Principal Is Named After Family Matters
Mark's high school principal is voiced by Reginald VelJohnson and, fittingly, the school itself is called Reginald VelJohnson High. His character is named B.N. Winslow, a direct nod to Carl Winslow, the role that made VelJohnson a household name on Family Matters. He's probably better known to some fans as the cop who helps John McClane in Die Hard, too.

Long before they were undead bosses guarding treasure in Morytania, the Barrows Brothers were six ordinary farmer siblings from a small village. It's a genuinely dark backstory too, tangled up in a love triangle, a jealous murder, and a drunken bar fight, before a mysterious stranger swooped in and offered them power in exchange for their loyalty.
